Action And Clinical Pharmacology: Deficiencies of one or more of the fat-soluble vitamins may occur in the presence of steatorrhea or other malabsorption, hepatic dysfunction, protein malnutrition, zinc deficiency, and very low fat diets. As with all vitamins, adequate levels are essential to good health and proper growth. Early nutritional intervention in disease such as cystic fibrosis appears crucial to growth and long-term outcome of these patients. Deficiencies lead to a wide variety of pathologies.
Vitamin A is essential for growth and bone development, vision, reproduction and maintenance of the integrity of mucosal and epithelial surfaces. Vitamin A deficiencies may lead to xerophthalmia, epithelial metaplasia of mucous membranes, decreased resistance to infection (characteristic change in epithelium of eye), night blindness and hyperkeratosis of the skin.
Vitamin D is essential for the absorption and utilization of calcium and phosphate and aids in the mobilization of bone calcium and maintenance of serum calcium concentrations.
Vitamin E is considered an essential element of human nutrition. Many of its actions are related to its antioxidant properties. Vitamin E deficiency rarely causes clinical symptoms in adults. In premature neonates, irritability, edema, thrombosis, and hemolytic anemia may be caused by vitamin E deficiency.
Vitamin K is necessary for synthesis in the liver of factor II (prothrombin), factor VII (proconvertin), factor IX (thromboplastin), and factor X. Deficiency of vitamin K or disturbances of liver function lead to deficiencies of these factors, with either latent or manifest hemorrhage. Vitamin K deficiency may occur in patients with biliary obstruction or other conditions limiting absorption of vitamin K such as celiac disease, ulcerative colitis, sprue, regional enteritis, cystic fibrosis, and intestinal resection and in patients receiving drugs that may affect liver function or intestinal flora.
Indications And Clinical Uses: To provide nutritional supplementation in individuals with malabsorptive conditions. ADEKs uses water- miscible forms of the fat-soluble vitamins to improve absorption in these individuals. It also provides vitamin C, B complex vitamins, and other vitamins plus zinc to supplement dietary intake.
For nutritional supplementation in individuals with deficient diets or difficulty in absorbing fat-soluble vitamins such as may occur in cystic fibrosis. For use solely under medical supervision.
Contra-Indications: None known.
Precautions: General: Do not exceed recommended dose. Pernicious anemia should be excluded before using this product since folic acid in doses above 0.1 mg daily may mask symptoms.
Pregnancy: Excessive amounts of vitamin A may be hazardous to the embryo or fetus when taken during pregnancy. Women of childbearing potential should consult their physicians regarding total vitamin A intake.
Anticoagulant Therapy: Vitamin K interferes with actions of anticoagulant drugs. Patients taking anticoagulants should consult their physicians before taking ADEKs.
Dosage And Administration: Pediatric Drops: 0 months to 1 year: 1 mL daily; 1 to 3 years: 2 mL daily, or as directed by physician. Intended for use solely under medical supervision. Shake well before each use.
Tablets: 4 to 10 years: 1 tablet daily; over 10 years: 2 tablets daily, or as directed by physician.
Tablets must be chewed or crushed thoroughly. Tablets may also be crushed and given with soft foods for children unable to chew tablets.
Availability And Storage: Pediatric Drops: Each mL contains: vitamin A (palmitate) 1 500 IU, vitamin D (cholecalciferol) 400 IU, vitamin E (d-a-tocopheryl) (vitamin E in water-miscible form as d-a-tocopheryl polyethylene glycol-1 000 succinate) 40 IU, vitamin K (phytonadione) 0.1 mg, vitamin C (ascorbic acid) 45 mg, thiamine (vitamin B1) 0.5 mg, riboflavin (vitamin B2) 0.6 mg, niacinamide 6 mg, vitamin B6 (pyridoxine HCl) 0.6 mg, vitamin B12 (cyanocobalamin) 4 g, biotin 15 g, pantothenic acid (d-pantothenol) 3 mg, zinc (as sulfate) 5 mg and beta carotene (source of vitamin A) 1 mg. Nonmedicinal ingredients: glycerin, natural and artificial flavors, propylene glycol, simethicone emulsion, sodium hydroxide, sodium saccharin and water. Bottles of 60 mL. Do not use if seal on cap is broken or missing. Keep out of reach of children. Store in a cool place away from direct light.
Tablets: Each tan tablet contains: vitamin A (palmitate) 4 000 IU, beta carotene (source of vitamin A) 3 mg, vitamin D (cholecalciferol) 400 IU, vitamin E (succinate) 150 IU, vitamin K (phytonadione) 0.15 mg, vitamin C (ascorbic acid) 60 mg, folic acid 0.2 mg, thiamine (vitamin B1) 1.2 mg, riboflavin (vitamin B2) 1.3 mg, niacin 10 mg, vitamin B6 (pyridoxine) 1.5 mg, vitamin B12 (cyanocobalamin) 12 g, biotin 50 g, pantothenic acid 10 mg and zinc (oxide) 7.5 mg. Nonmedicinal ingredients: dextrose with corn syrup solids, flavor, fructose, glycirrhizic acid, magnesium stearate, silicon dioxide and stearic acid. Contains no dyes or artificial sweeteners. Bottles of 60. Dispense in tight, light-resistant containers.
